這三種RTX-毒素,ApxI、Apx-II和Apx-III分別具有顯著的溶血活性(ApxI)、溫和的溶血活性(Apx-II)或巨噬細(xì)胞的細(xì)胞毒活性(Apx-III)。
各種毒素的活性以不同的血清型相當(dāng)隨機(jī)地劃分。存在四種亞型由血清型1,5,9和11所代表的亞型A,產(chǎn)生ApxI和Apx-II,由血清型2,3,4,6和8所代表的亞型B,產(chǎn)生Apx-II和Apx-III,由血清型10所代表的亞型C,僅產(chǎn)生ApxI,由血清型7和12所代表的亞型D,僅產(chǎn)生Apx-II,已知,ApxI、-II和-III是抗胸膜肺炎放線桿菌感染的一般疫苗中的必需成分不包括至少ApxI、-II和III的疫苗將不提供對(duì)所有胸膜肺炎放線桿菌血清型的保護(hù)。同樣,不包括至少一種特異血清型Apx-毒素的疫苗將甚至不會(huì)誘導(dǎo)對(duì)單-血清型的保護(hù)。
較早時(shí)期已描述了基于體外合成,喪失其毒性的胸膜肺炎放線桿菌RTX-毒素的亞單位疫苗,如在歐洲專利EP NO.0354.628中所述,其中公開(kāi)了基于胸膜肺炎放線桿菌溶血素和細(xì)胞毒素的亞單位疫苗,并且在歐洲專利EP NO.0453.024中所述,其中公開(kāi)了基于溶血素、細(xì)胞毒素和外膜蛋白的胸膜肺炎放線桿菌亞單位疫苗。
然而對(duì)亞單位疫苗來(lái)說(shuō)一般具有4個(gè)重要的不足·為了充分地激活免疫系統(tǒng)需要大量的免疫原性材料。
·通常僅B-細(xì)胞免疫被激活。
·幾種保護(hù)性抗原僅在體內(nèi)被激活,且因此不能夠以亞單位疫苗形式存在。
·活的病原性細(xì)菌具有許多重要的免疫原性分子,如外膜蛋白和莢膜蛋白,它們均對(duì)于保護(hù)至關(guān)重要且因此被包括進(jìn)有效的亞單位疫苗中。
僅次于上述第1和第2點(diǎn),尤其是第4點(diǎn)的明顯問(wèn)題使得難以制備有效的亞單位疫苗。
這由例如在上述歐洲專利EP NO.0.453.024中公開(kāi)的胸膜肺炎放線桿菌亞單位疫苗加以說(shuō)明,其中4種不同的亞單位(三種RTX-毒素和外膜蛋白)結(jié)合于一種疫苗中。
很顯然,為了克服抗Pasteurellaeeae感染亞單位疫苗的不足,一種活的減毒疫苗將是高度需要的。
活減毒疫苗具有下面的優(yōu)勢(shì)其可用低劑量施用(其是自我復(fù)制的)其密切模擬天然/野生-型感染其同時(shí)提供所有可能的免疫學(xué)重要的抗原。不過(guò),盡管明顯的優(yōu)勢(shì),本發(fā)明之前還沒(méi)有商業(yè)上可得到的基于胸膜肺炎放線桿菌的活疫苗。其原因在于下面似是而非的論點(diǎn)如前述,Apx-I、-II和-III均是抗胸膜肺炎放線桿菌感染普通疫苗的必需成分。因此活疫苗必需產(chǎn)生這三種RTX-毒素。然而這三種RTX-毒素是所有放線桿菌種類中強(qiáng)有力的毒性因子(見(jiàn),如Coote,J.G.;FEMS微生物學(xué)綜述88137-162(1992)),Tascon等.;分子微生物學(xué),14207-216(1994),Jansen等.;感染與免疫.6327-37(1995))。
為了減弱活A(yù)pp菌株的毒力而對(duì)RTX-毒素的缺失在技術(shù)上是可行的,但其沒(méi)有提供解決問(wèn)題的途徑由于它們不再在宿主中誘導(dǎo)對(duì)胸膜肺炎放線桿菌溶血/細(xì)胞毒活性的免疫性,故這種RTX-陰性菌株作為活減毒疫苗菌株將是無(wú)用的。
然而到現(xiàn)在,盡管在誘導(dǎo)免疫性中是重要的,但在建立免疫性中較ApxI、-II和-III是次要的毒力因子還是未知的。
因此,在App基因組中具有這樣的位點(diǎn)是高度必要的,即該位點(diǎn)當(dāng)被修飾時(shí)有利于毒力且因此導(dǎo)致減毒的App菌株,而同時(shí),盡管其在激發(fā)免疫性中是有用的,但從疫苗觀點(diǎn)來(lái)看是必不可少的。然而這樣的位點(diǎn)目前還是未知的。此外,如果這樣的位點(diǎn)將普遍存在于所有的App菌株中而不是局限于某些血清型中,該位點(diǎn)是高度必要的。那么這樣的位點(diǎn)將通過(guò)相同位點(diǎn)的缺失允許對(duì)所有不同的血清型減毒。
提供普遍存在于無(wú)論什么血清型的所有胸膜肺炎放線桿菌菌株中的這種減毒位點(diǎn)是本發(fā)明的目的之一。
最近,在胸膜肺炎放線桿菌血清型1菌株中發(fā)現(xiàn)了一種新的基因(論文T.J.Anderson Nov.1995)。
盡管該基因不象已知的放線桿菌的ApxI、-II和-III基因,但其與已知的腦膜炎奈氏球菌的RTX-毒素具有相似性,正是由于此原因?qū)⑵涿麨镽TX-基因apxIV。然而該基因幾乎在所有的方面均與上述存在于各種放線桿菌科的不同種中的三種已知RTX-毒素基因apxI、-II和-III不同。首先,基因組結(jié)構(gòu)完全不同。其次,沒(méi)有如同發(fā)現(xiàn)于已知Apx-毒素的激活機(jī)制。第三,在那時(shí)沒(méi)有特異性的體內(nèi)溶血或細(xì)胞毒活性可歸結(jié)于該基因或其可能的基因產(chǎn)物。
現(xiàn)已驚奇地發(fā)現(xiàn)該基因與三種已知的RTX-基因完全不同,無(wú)論其什么血清型存在于胸膜肺炎放線桿菌的所有種類中。這通過(guò)探針雜交加以測(cè)定,該探針包括apxIV編碼序列,此序列中具有實(shí)施例6和7中所述所有血清型胸膜肺炎放線桿菌的DNA限制性片段。出乎意料的是現(xiàn)已發(fā)現(xiàn),雖然apxIV缺失突變體是存活的,但與其具有apxIV的母本菌株相比,它們毒性較小。

有功能的ApxIV毒素被認(rèn)為是這樣的一種蛋白即其具有在野生型細(xì)菌中表達(dá)ApxIV毒素的所有特征,并且以野生型水平表達(dá)。因此,無(wú)功能的ApxIV毒素被認(rèn)為是這樣的一種毒素即其缺乏在野生型細(xì)菌中表達(dá)ApxIV毒素的部分或所有特征,并且/或者以不足以獲得ApxIV毒素野生型效應(yīng)的水平表達(dá)。
產(chǎn)生ApxIV毒素能力的喪失可,例如是由于編碼ApxIV毒素編碼序列中的修飾。其也可是由于,例如,已知涉及apxIV基因轉(zhuǎn)錄的區(qū)域,如啟動(dòng)子區(qū)域的修飾或由于涉及翻譯區(qū)域,如核糖體結(jié)合位點(diǎn)的修飾。
apxIV基因的整體結(jié)構(gòu)列于圖1中。
該圖中,ApxIV毒素特征性的同向重復(fù)區(qū)由陰影框(dashed box)表示,而也是ApxIV特異的富含-甘氨酸的非肽(nonapeptide)區(qū)由黑箭頭表示。該重復(fù)區(qū)發(fā)現(xiàn)于ApxIV的C-末端部分。這些鑒別性特征存在于所有的胸膜肺炎放線桿菌血清型中。兩種血清型的核苷酸序列和氨基酸序列列于SEQ.ID NO.1-4中。SEQ.ID.NO.1顯示了App血清型1apxIV基因的核酸序列,且SEQ.ID.NO.2顯示了血清型1ApxIV毒素的相應(yīng)氨基酸序列。SEQ.ID.NO.3顯示了App血清型3apxIV基因的核苷酸序列,而SEQ.ID.NO.4顯示了血清型3ApxIV毒素的相應(yīng)氨基酸序列。圖2顯示各種胸膜肺炎放線桿菌血清型的Apx-毒素之間的氨基酸水平,特別位于N-末端的650個(gè)氨基酸具有相當(dāng)高水平的保守性。這也是apxIV基因的異常特性。從圖1中顯而易見(jiàn),根據(jù)血清型的不同,位于該毒素C-末端重復(fù)數(shù)目的差異可能發(fā)生。此差異解釋了獲自各種血清型基因及編碼毒素大小的差異。
甚至在分離自相同血清型的胸膜肺炎放線桿菌不同分離物的apxIV基因之間可能有某些核苷酸序列的差異。這是由于本領(lǐng)域眾所周知的存在于所有有機(jī)體中的自然變異。在由apxIV基因編碼的ApxIV毒素中某些氨基酸由其它血清型ApxIV毒素中的別的氨基酸所替代是可能的,然而該多肽功能沒(méi)有改變。例如,在某一位點(diǎn)含有Asp的多肽和其相應(yīng)位點(diǎn)含有Asn的變異體仍然具有相同的特性。其中的氨基酸由功能類似氨基酸所替代的方法稱為功能性替換。此時(shí)的變異體蛋白稱為功能性變異體。另一種變異的原因是遺傳密碼簡(jiǎn)并現(xiàn)象。簡(jiǎn)言之,其意思為,例如氨基酸谷氨酸由GAT和GAA所編碼。除了Met和Trp以外,此現(xiàn)象適用于所有的氨基酸。因此,很明顯,例如本發(fā)明給出的血清型1ApxIV毒素可不僅由SEQ ID NO1中的核苷酸序列所編碼而且也可由很多種其它序列所編碼,所有的仍然得到相同或功能相同的多肽。
因此,編碼與ApxIV毒素功能相當(dāng)多肽的變異apxIV序列屬于本發(fā)明的范圍之內(nèi)。
根據(jù)本發(fā)明的活減毒細(xì)菌可以幾種方式獲得。獲得該菌一種可能的方式是通過(guò)經(jīng)典方法的途徑,如以誘變劑如堿基類似物處理野生型胸膜肺炎放線桿菌,以紫外光處理或溫度處理細(xì)菌。不產(chǎn)生有功能ApxIV毒素的菌株不誘導(dǎo)或以較小程度誘導(dǎo)抗ApxIV毒素抗體,且因此可容易地在動(dòng)物測(cè)試中加以篩選。必需的抗血清可按下面實(shí)施例3中描述獲得。另一種可能性是用重組DNA-技術(shù)有意識(shí)地向編碼ApxIV毒素的基因中導(dǎo)入詳細(xì)定義的突變。這樣的突變可以是插入、缺失、一個(gè)核苷酸被另一個(gè)所替代或其組合,唯一的條件是該突變基因不再編碼有功能的ApxIV毒素??扇菀椎乜闯觯绕涫窃陂_(kāi)放閱讀框中導(dǎo)入終止密碼子的突變,或?qū)е麻_(kāi)放閱讀框中移碼的突變很適于獲得不再編碼有功能ApxIV毒素的菌株。這種突變可通過(guò),例如,用Clontech銷售的Transformer試劑盒的體外定點(diǎn)誘變方法加以完成。許多其它標(biāo)準(zhǔn)的重組DNA技術(shù)如以限制性酶消化基因,然后以內(nèi)切核酸酶處理和重新連接同樣是可用的。
因此,在優(yōu)選的方式中,本發(fā)明的該實(shí)施方案涉及其中編碼ApxIV毒素的基因包括突變的活減毒細(xì)菌。
包括apxIV基因片段或甚至整個(gè)基因缺失,或異源DNA-片段插入的詳細(xì)定義的突變,與經(jīng)典誘導(dǎo)的突變相比,具有這樣的優(yōu)勢(shì)即它們將不回復(fù)到野生型狀況。
因此,在更為優(yōu)選的方式中,本發(fā)明的該實(shí)施方案指其中編碼ApxIV毒素的基因包括插入和/或缺失的活減毒細(xì)菌。
如果現(xiàn)將大量的疫苗施于豬中,只要是為了降低接種費(fèi)用的原因,很明顯幾種疫苗的聯(lián)合施用將會(huì)是合乎需要的。因此,使用活體減毒疫苗菌株作為編碼選自其它病原性微生物或病毒抗原的異源基因的重組載體是很有吸引力的。施用這樣的重組載體具有這樣的優(yōu)勢(shì),即施用該載體后,誘導(dǎo)出同時(shí)抗兩種或更多種疾病的免疫性。因?yàn)榫幋aApxIV毒素的基因可用作這樣異源基因的插入位點(diǎn),所以根據(jù)本發(fā)明的活減毒細(xì)菌提供了異源基因很適宜的載體。ApxIV基因用作插入位點(diǎn)具有這樣的優(yōu)勢(shì)即apxIV基因被同時(shí)失活,并且新導(dǎo)入的異源基因可根據(jù)同源的胸膜肺炎放線桿菌基因來(lái)表達(dá)。這種重組載體的構(gòu)建可用標(biāo)準(zhǔn)的分子生物學(xué)技術(shù)如同源重組按常規(guī)地加以完成。因此,在甚至更為優(yōu)選的實(shí)施方案中,本發(fā)明涉及這樣的活減毒的胸膜肺炎放線桿菌即其不產(chǎn)生有功能的ApxIV毒素,并且其中有異源基因插入到ApxIV基因中。如上述,這樣的異源基因可以是例如,編碼選自其它病原微生物或病毒抗原的基因。另一種可能性是插入編碼涉及激發(fā)免疫系統(tǒng)的蛋白,如白細(xì)胞介素或干擾素的基因。
在本發(fā)明更為優(yōu)選的形式中,異源基因編碼一個(gè)或更多個(gè)選自以下類群的抗原,包括豬生殖呼吸綜合癥(PRRS)病毒、假狂犬病毒、豬流感病毒、豬細(xì)小病毒、傳染性胃腸炎病毒(Transmissible Gastroenteritisvires)、輪狀病毒、大腸桿菌、豬丹毒絲菌、多殺巴斯德氏菌、支氣管炎博德特氏菌、副豬嗜血菌及豬鏈球菌。

在自然狀態(tài)下控制apxIV基因表達(dá)的可開(kāi)關(guān)啟動(dòng)子現(xiàn)被發(fā)現(xiàn)位于SEQID NO5中位置451和1132之間的DNA片段中。很明顯該DNA片段中那些非啟動(dòng)子必需成分的部分不必要存在于該片段中。因此,保留該啟動(dòng)子活性的此DNA片段的較短部分同樣適用于異源基因的表達(dá)。因此,該實(shí)施方案更為優(yōu)選的形式涉及包括SEQ ID NO5中位置451到1132DNA片段或其仍具有啟動(dòng)子活性子片段的核苷酸序列。
細(xì)菌啟動(dòng)子均具有兩個(gè)共有區(qū),所謂的-10和-35區(qū)。雖然這些共有區(qū)的側(cè)翼序列可能在一定程度上影響啟動(dòng)子的效率,僅使用包括-35與ATG密碼子之間的DNA片段的啟動(dòng)子區(qū)域部分可能是優(yōu)越的。該DNA片段位于SEQ ID NO5的位置617與位置641之間。因此,在本發(fā)明該實(shí)施方案更為優(yōu)選的形式中涉及包括SEQ ID NO5中位置617至641之間DNA片段的核苷酸序列。
本發(fā)明也涉及作為亞單位疫苗成分的ApxIV毒素。亞單位疫苗將最為可能包括這三種已知的Apx-毒素。這一點(diǎn)上面已經(jīng)提到。然而,如上述,由于意外地發(fā)現(xiàn)ApsIV毒素存在于所有胸膜肺炎放線桿菌血清型中,故其是亞單位疫苗理想的添加成分產(chǎn)生的抗ApxIV毒素的中和抗體提供了對(duì)無(wú)論什么血清型的每種及所有胸膜肺炎放線桿菌株所產(chǎn)生ApxIV毒素的保護(hù)。因此,本發(fā)明的另一個(gè)實(shí)施方案涉及用于保護(hù)動(dòng)物免受胸膜肺炎放線桿菌細(xì)菌感染的亞單位疫苗,其中包括純化的ApxIV毒素。ApxIV毒素可單獨(dú)施用,或與任何或所有上述的毒素ApxI、-II和-III結(jié)合施用和/或,例如,與胸膜肺炎放線桿菌外膜蛋白(OMPs)結(jié)合施用??赏ㄟ^(guò)將ApxIV毒素以足以誘導(dǎo)免疫反應(yīng)的量與藥物上可接受的載體混合而容易地制備該疫苗。通過(guò)在適當(dāng)?shù)谋磉_(dá)載體中導(dǎo)入apxIV基因、表達(dá)該基因及分離毒素而制備ApxIV毒素是可能的。本領(lǐng)域中已知有多種通用表達(dá)系統(tǒng),如細(xì)胞表達(dá)系統(tǒng),桿狀病毒表達(dá)系統(tǒng)及哺乳動(dòng)物細(xì)胞表達(dá)系統(tǒng)。在實(shí)施例3中描述了如何通過(guò)大腸桿菌中表達(dá)該基因而獲得ApxIV毒素。
本發(fā)明的另一實(shí)施方案涉及活減毒疫苗,其中包括上述用于保護(hù)動(dòng)物免受胸膜肺炎放線桿菌感染的活減毒細(xì)菌。這種疫苗可通過(guò)將活減毒細(xì)菌與藥物上可接受的載體混合而獲得。這些疫苗包括至少免疫原性有效量的根據(jù)本發(fā)明活減毒產(chǎn)生菌。免疫原性有效量意思為接種時(shí)施用的活減毒菌的量足以在宿主內(nèi)誘導(dǎo)對(duì)產(chǎn)生毒性形式RTX-毒素菌的有效免疫反應(yīng)。待施用的有用劑量將隨著年齡、體重及接種的動(dòng)物,施用的方式和接種所要針對(duì)病原體的類型而改變。此疫苗包括足以導(dǎo)致免疫反應(yīng)的任何劑量的細(xì)菌。例如,103與105個(gè)細(xì)菌之間的劑量范圍是很合適的劑量。
藥物上可接受的載體可以是簡(jiǎn)單的如水,但其也可以包括,例如,在其中培養(yǎng)細(xì)菌的培養(yǎng)液。其它合適的載體例如,生理鹽濃度的溶液。用于本發(fā)明其它藥物上可接受載體或稀釋劑的實(shí)例包括穩(wěn)定劑如SPGA、碳水化合物(如山梨醇、甘露醇、淀粉、蔗糖、葡萄糖、葡聚糖)、蛋白如清蛋白或酪蛋白、含有蛋白如牛血清或去脂牛奶的試劑及緩沖液(如磷酸鹽緩沖液)。
任選地,具有佐劑活性的一種或多種化合物可加入到疫苗中。佐劑為非特異性的免疫系統(tǒng)刺激劑。它們?cè)鰪?qiáng)宿主對(duì)入侵病原體的免疫反應(yīng)。本領(lǐng)域已知佐劑的實(shí)例有弗氏完全和不完全佐劑,維生素E,非離子型嵌段聚合物,胞壁酰二肽,ISCOMs(免疫刺激復(fù)合物,參考如歐洲專利EP109942),皂角苷,礦物油,植物油和Carbopol(均聚物)。特別是適用于粘膜應(yīng)用的佐劑有,例如,大腸桿菌熱不穩(wěn)定性毒素(LT)或霍亂毒素(CT)。
其它合適的佐劑有,例如,氫氧化鋁,磷酸鋁或氧化鋁,油性乳劑(如Bayol F(R)或Marcol52(R),皂角苷或維生素E-加溶物。
因此,在優(yōu)選形式中,根據(jù)本發(fā)明的疫苗包括佐劑。
為了施用到動(dòng)物,根據(jù)本發(fā)明的疫苗可特別地以鼻內(nèi)、皮內(nèi)、皮下、通過(guò)煙霧劑或肌肉內(nèi)施用。
有幾種保存亞單位和活生物的方式。例如在冰箱內(nèi)保存是眾所周知的方法。在含有甘油的緩沖液中于-70℃保存也常使用。細(xì)菌也可保存于液氮中。冷凍干燥是另一種保存方式。冷凍干燥的細(xì)菌可被保存并維持存活多年。冷凍干燥的細(xì)菌的保存溫度可位于零度以上而不損害其存活力。冷凍干燥同樣適用于亞單位。
冷凍干燥可根據(jù)眾所周知的標(biāo)準(zhǔn)冷凍干燥方法加以完成。任選有利的添加劑,如去脂牛奶、海藻糖、明膠或牛血清白蛋白可在冷凍過(guò)程中加入。
因此,在更為優(yōu)選的實(shí)施方案中,根據(jù)本發(fā)明的疫苗是冷凍的形式。
在該實(shí)施方案更為優(yōu)選的形式中,疫苗還額外包括一種或多種選自其它病原體微生物或病毒的抗原。這種疫苗可通過(guò)將選自其它病原菌或病毒的一種或多種抗原添加到根據(jù)本發(fā)明的活減毒菌及上述藥物上可接受的載體中而獲得。
當(dāng)然,不僅添加一種或多種抗原,而且添加一種或多種整個(gè)病原體如,失活或活性形式病原體是可能的。
作為選擇,可用上述已知的重組DNA技術(shù)將編碼一種或多種選自其它病原微生物或病毒的抗原的遺傳信息克隆到活減毒菌中而獲得疫苗。無(wú)論從醫(yī)學(xué)還是從身體角度看,這種疫苗當(dāng)然較以每種病原體單獨(dú)接種對(duì)接種的動(dòng)物產(chǎn)生較小的協(xié)迫。
在甚至更為優(yōu)選的形式中,這些抗原選自豬生殖呼吸綜合癥(PRRS)病毒、偽狂犬病毒、豬流感病毒、豬細(xì)小病毒、傳染性胃腸炎病毒、輪狀病毒、大腸桿菌、豬丹毒絲菌、多殺巴斯德氏菌、支氣管炎博德特氏菌、副豬嗜血菌及豬鏈球菌。
本發(fā)明也涉及不能產(chǎn)生有功能ApxIV毒素的各種活減毒胸膜肺炎放線桿菌的制備方法。這些方法包括在編碼apxIV蛋白的基因中導(dǎo)入突變。用誘變劑的經(jīng)典突變技術(shù)和本領(lǐng)域眾所周知的用于apxIV基因遺傳信息插入、替代或缺失的重組DNA技術(shù)是可用的。
在更為優(yōu)選的形式中,上述方法用于導(dǎo)入缺失。
包括將根據(jù)本發(fā)明的細(xì)菌與藥物上可接受的載體混合的用于本發(fā)明活減毒疫苗制備的方法也是本發(fā)明的一部分。
用于亞單位疫苗制備的方法也在本發(fā)明的范圍之內(nèi)。該方法包括將純化的ApxIV毒素與藥物上可接受的載體混合。
在活疫苗接種領(lǐng)域中另一個(gè)普遍認(rèn)識(shí)到的問(wèn)題如下在宿主動(dòng)物血清中抗某種病原體抗體的存在表明該宿主已被此毒性或減毒形式的病原體感染。然而區(qū)分野外感染的動(dòng)物與以活疫苗菌株接種的動(dòng)物是不可能的。根據(jù)本發(fā)明的活體減毒胸膜肺炎放線桿菌按如下提供了該問(wèn)題的解決方案如在實(shí)施例3中所述,胸膜肺炎放線桿菌血清型1的apxIV基因已被分離并在異源宿主細(xì)胞中表達(dá)。將此表達(dá)產(chǎn)物進(jìn)行PAAGE-凝膠電泳且然后用于Western-印跡。將印跡與獲自以胸膜肺炎放線桿菌有意識(shí)感染豬的恢復(fù)期血清及獲自野生菌株感染的血清孵育。發(fā)現(xiàn)apxIV基因在所有測(cè)試的胸膜肺炎放線桿菌野生菌株中均有體內(nèi)表達(dá)。這表明以胸膜肺炎放線桿菌感染的豬將一直具有抗所有感染菌株血清型的感染它們的菌株的抗體。由于apxIV基因的缺失,根據(jù)本發(fā)明的活減毒菌不再產(chǎn)生ApxIV毒素。因此以本發(fā)明的活減毒胸膜肺炎放線桿菌菌株接種的動(dòng)物在其血清中將不具有抗ApxIV毒素的抗體。
在比較測(cè)試,如ELISA測(cè)試中,該血清將因此與所有免疫原性的胸膜肺炎放線桿菌-蛋白如ApxI、II和/或III反應(yīng),但將不與ApxIV起反應(yīng)。然而,以胸膜肺炎放線桿菌野生菌株感染豬的血清將與所有免疫原性的胸膜肺炎放線桿菌-蛋白,包括ApxIV起反應(yīng)。因此,根據(jù)本發(fā)明的活減毒胸膜肺炎放線桿菌證明是很適宜的標(biāo)記疫苗,即可與野生菌株區(qū)分的疫苗菌株。
用于區(qū)分疫苗菌株與野生菌株的診斷測(cè)試可以是簡(jiǎn)單的ELISA-測(cè)試,其中的純化ApxIV毒素涂到ELISA-平板的孔壁上。然后與待測(cè)的豬血清孵育,接著與,如標(biāo)記的抗豬抗體孵育可顯示抗ApxIV毒素的抗體存在與否。
另一種診斷測(cè)試系統(tǒng)的實(shí)例是,如,將包括純化ApxIV毒素的Western印跡與待測(cè)的豬血清孵育,接著檢測(cè)特異性的抗-ApxIV抗體。
因此,用于區(qū)分以胸膜肺炎放線桿菌野生菌株感染豬的血清與以包括本發(fā)明活減毒疫苗胸膜肺炎放線桿菌菌株疫苗接種豬的血清(包括純化ApxIV毒素)的診斷測(cè)試也在本發(fā)明的范圍之內(nèi)。
豬健康保護(hù)中的另一問(wèn)題如下即難以用快捷而清晰的方式測(cè)定豬是否以胸膜肺炎放線桿菌或豬放線桿菌感染,或是二者共同感染。目前還沒(méi)有用于對(duì)豬放線桿菌特異性檢測(cè)的診斷測(cè)試。這主要是由于這樣的事實(shí),即胸膜肺炎放線桿菌與豬放線桿菌共有許多相同的抗原。兩種高度抗原性的Apx-毒素可作為實(shí)例;在如豬放線桿菌中,ApxI與ApxII具有高度保守的同源物(Van Ostaayen等.,待發(fā)表)。
編碼ApxI、-II和-III毒素或同源物的已知RTX基因?qū)嶋H上發(fā)現(xiàn)于放線桿菌屬的所有成員中,如胸膜肺炎放線桿菌、豬放線桿菌、羅氏放線桿菌及馬駒放線桿菌。因此,本發(fā)明者最初認(rèn)為,新的RTX-毒素ApxIV在所有放線桿菌屬成員中是相同的。然而,測(cè)試豬病原性放線桿菌后發(fā)現(xiàn),又一次與已知三種RTX-基因驚奇地形成鮮明對(duì)照,即該新的RTX-基因apxIV僅存在于豬病原體胸膜肺炎放線桿菌中。其沒(méi)有出現(xiàn)于所有其它常見(jiàn)的豬病原性放線桿菌各種類中,且因此它也沒(méi)有出現(xiàn)于豬放線桿菌中。見(jiàn)實(shí)施例6和7。因此,令人驚奇地注意到豬血清中抗ApxIV抗體的存在是豬被胸膜肺炎放線桿菌感染而不是被豬放線桿菌或任何其它豬病原體放線桿菌種類感染的快捷而又清晰的證據(jù)。
這樣,本發(fā)明也提供了基于抗ApxIV抗體存在與否的診斷測(cè)試,且因此用于特異性區(qū)分胸膜肺炎放線桿菌感染與豬放線桿菌感染的區(qū)別測(cè)試。這種測(cè)試可以是,如,ELISA測(cè)試,其在分離的小孔中包括既存在于胸膜肺炎放線桿菌又存在于豬放線桿菌中的ApxI及-II毒素和純化的ApxIV毒素。豬放線桿菌感染的動(dòng)物血清將僅與包括ApxI和Apx-II的小孔起反應(yīng)而胸膜肺炎放線桿菌感染的動(dòng)物也將與包括純化ApxIV毒素的小孔起反應(yīng)。
實(shí)施例1胸膜肺炎放線桿菌血清型1apxIV基因的克隆與分析標(biāo)準(zhǔn)的分子生物學(xué)方法(質(zhì)粒DNA分離,限制性酶切、瓊脂糖凝膠電泳、Southern印跡、連接、轉(zhuǎn)化、電穿孔),如果沒(méi)有另作說(shuō)明,基本上按Sambrook等(分子克隆,實(shí)驗(yàn)手冊(cè),冷泉港、紐約,1989)或Ausubel等(當(dāng)前分子生物學(xué)方法,John Wiley &Sons,紐約,1987)的描述進(jìn)行。PCR基本上按Innis等(PCR方法,方法及應(yīng)用指南,學(xué)術(shù)出版公司,圣地亞哥,1990)所述進(jìn)行。染色體DNA分離根據(jù)Pitcher等(應(yīng)用微生物學(xué)通迅8;151-156,1989)完成。所有胸膜肺炎放線桿菌參比菌株(血清型1菌株4074;血清型2菌株S1536血清型3S1421;血清型4M62;血清型5aK17;血清型5bL20;血清型6femφ;血清型7WF83;血清型8405;血清型9CVI13261;血清型1013039;血清型1156153及血清型128329)的來(lái)源由Frey和Nicolet描述(臨床微生物雜志,28;232-236,1990)。胸膜肺炎放線桿菌血清型3菌株HV114為野生型分離子(即在Beck等,臨床微生物學(xué)雜志.,32;2749-2754,1994中測(cè)試的血清型3菌株之一)。所用的其它放線桿菌菌株;羅氏放線桿菌ATCC27072;馬駒放線桿菌ATCC19392;豬放線桿菌ATCC15558。溶血巴斯德氏菌1型菌株ATCC14003被使用。
所用的大腸桿菌宿主菌株XL1-blue(Stratagene,La Yolla,Ca;基因型rec A1 end A1 gyrA96 thi-1 hsd R17 sup E44 rel A 1lac[F’proABlaclqZDM 15 Tn10(Tetr)])及HMS174(DE3)(AMS生物技術(shù)公司,瑞士;基因型F-rec A(rk12-mk12+)rifrIDE3)。根據(jù)獲自T.J.,Anderson論文的初步序列數(shù)據(jù)(Guelph大學(xué),1995),合成兩條命名為APXIVA-1L(5’-TGGCACTGACGGTGATGA-3’)和APXIVA-1R(5’-GGCCATCGACTCAACCAT-3’)的引物。這些引物用于以胸膜肺炎放線桿菌血清型3菌株HV114和血清型1參照菌株4074的染色體DNA作為模板的PCR擴(kuò)增中。用兩種菌株擴(kuò)增出442bp的片段。將獲自血清型3染色體DNA的此片段根據(jù)制造商方法手冊(cè)以洋地黃毒苷-11-dUTP(寶靈曼)標(biāo)記(此片段命名為探針APXIVA,見(jiàn)圖4)。隨后將該標(biāo)記探針用于與含有ClaI消化的菌株4074染色體DNA的Southern印跡雜交。該探針與大約8.0kb的片段雜交。血清型1菌株4074的apxIV基因通過(guò)將ClaI消化的染色體DNA連接到ClaI消化的pBluescript II SK-(Stratagene美國(guó))中而加以分離。以連接的DNA轉(zhuǎn)化大腸桿菌菌株XL1-blue并將轉(zhuǎn)化體于含有100mg/ml氨芐青霉素的LB平板上進(jìn)行篩選。
用APXIVA探針通過(guò)平板硝酸纖維素復(fù)制品的菌落雜交篩選帶有apxIV的克隆。這樣,分離到命名為pROK7的質(zhì)粒并顯示其帶有約8kb的ClaI插入子。將ClaI插入子的頭6736bp進(jìn)行測(cè)序(SEQ ID1)并鑒定出編碼具有約186KD預(yù)期大小的編碼1657個(gè)氨基酸殘基蛋白(SEQID2)的4971個(gè)核苷酸的開(kāi)放閱讀框。該基因命名為apxIV-var1(見(jiàn)圖3)。
實(shí)施例2胸膜肺炎放線桿菌血清型3apxIV基因的克隆與分析將標(biāo)記的探針APXIVA(在實(shí)施例1中提到)用于與含有ClaI消化的HV114菌株染色體DNA的Southern印跡雜交。探針與約7.0kb的片段雜交。將從HV114分離的染色體DNA以ClaI消化,并與ClaI消化的pBluescript II SK-(Stratagene美國(guó))連接。以連接的DNA轉(zhuǎn)化大腸桿菌菌株XL1-blue并于具有100mg/ml氨芐青霉素的LB平板上篩選轉(zhuǎn)化體。用APXIVA探針通過(guò)平板的硝酸纖維素復(fù)制品的菌落雜交篩選帶有apxIV的克隆。因此,分離出一個(gè)名為pROK5的質(zhì)粒并顯示其帶有約7kb的ClaI插入子。通過(guò)序列分析分析該插入子(SEQ ID 3)。鑒定出一個(gè)4146bp的開(kāi)放閱讀框,其編碼具有約154KD預(yù)期大小的1382氨基酸殘基的蛋白(SEQ ID 4)。該基因命名為apxIV-var3(見(jiàn)圖3)。
實(shí)施例3ApxIV var3-多聚組氨酸融合蛋白在大腸桿菌中的表達(dá)由質(zhì)粒pROK5,制備一個(gè)缺失克隆,其中含有apxIV基因的3’末端,從Bam HI位點(diǎn)(SEQ ID NO3中核苷酸NO.2747)到apxIV基因插入下游末端的ClaI位點(diǎn)。將該質(zhì)粒命名為pROK1。用寡核苷酸APXIVAHIS1-L(5’-AGCCATATGGGCGATTTAAATTTCAG-3’)及APXIVAHIS1-R(5’-TATGGATCCTCCGTGCTTCTGAGC-3’)和質(zhì)粒pROK1 DNA作為模板,擴(kuò)增出2.1kb的DNA片段(見(jiàn)圖4),其中含有分別位于5’和3’末端NdeI及BamHI限制性位點(diǎn)兩側(cè)的apxIV-var3(SEQID3)中3520到5643bp的區(qū)域。在表達(dá)載體pETHIS-1中克隆NdeI/BamHI消化的PCR片段后,以相同的酶消化,獲得名為pJFFapxIV6/10 his-1的質(zhì)粒。質(zhì)粒pETHIS-1是pET14b(Novagen公司.,Madison,Wi)的衍生物,其中的多克隆位點(diǎn)已被擴(kuò)展且已插入編碼組氨酸十聚體(decamer)的區(qū)域。因而,pJFFapxIV6/10 his-1質(zhì)粒含有T7啟動(dòng)子控制下的翻譯融合體,其編碼組氨酸六聚體、后接SEQ ID4中653到1360氨基酸殘基,后面再接上組氨酸十聚體。將此質(zhì)粒轉(zhuǎn)化至具有pLysS的大腸桿菌菌株HMS174(DE3)中,其中含有IPTG可誘導(dǎo)的T7RNA聚合酶基因和用于增加穩(wěn)定性的T7溶菌酶基因。將該菌株培養(yǎng)于含25mg/ml氯霉素及100mg/ml的氨芐青霉素的LB培養(yǎng)基至0.5的OD650,并以10mM濃度的異丙基-b-D-硫代吡喃半乳糖苷誘導(dǎo)。添加IPTG以后,細(xì)胞于37℃孵育2.5小時(shí),離心收獲細(xì)胞,并且分離包涵體形式的80kD預(yù)期大小的融合蛋白。將包涵體溶解于pH8.0的6M鹽酸胍、300mM NaCl和50mMNaH2PO4溶液中且80kD融合蛋白通過(guò)用Ni2+螯合柱(Qiagen AG,Basel)的固定金屬親和層析(IMAC)(Schmitt等,分子生物學(xué)報(bào)告18;223-230,1993)進(jìn)一步加以純化。以pH5.0從柱中洗脫純凈蛋白。將收集的餾份于pH8.0的300mM NaCl和50mM NaH2PO4溶液中透析。以用1體積完全弗氏佐劑(Difco實(shí)驗(yàn)室,Detroit,MI)混合的500mg多聚組氨酸融合產(chǎn)物免疫兔子。3周后施以混合有不完全弗氏佐劑的相同量的增強(qiáng)劑量。增強(qiáng)免疫后四周,將兔子放血并獲得包括抗-ApxIV毒素抗體稱為血清522-409的超免疫血清。
實(shí)施例4apxIV基因在體外培養(yǎng)的胸膜肺炎放線桿菌中的表達(dá)將血清型1的胸膜肺炎放線桿菌參比菌株培養(yǎng)于補(bǔ)充以10mg/ml b-NAD的Columbia肉湯中并按所述(Beck等,臨床微生物雜志,32;2749-2754,1994)進(jìn)行收獲。在相鄰于包括ApxIA、ApxIIA和ApxIVA-多聚組氨酸融合蛋白道的位置通過(guò)聚丙烯酰胺凝膠電泳(Laemmli,自然227680-685,1970)分離濃縮的培養(yǎng)上清并進(jìn)行Western印跡方法(Towbin等,美國(guó)自然科學(xué)院院報(bào)764350-4354,1979)。將Western印跡按Beek等所述(臨床微生物學(xué)雜志,32;2749-2754,1994)與抗-ApxIA-和抗-ApxIIA單克隆抗體及抗-ApxIV血清522-409(見(jiàn)實(shí)施例3)起反應(yīng)。分離的血清型1RTX毒素餾份清楚地含有ApxIA和ApxIIA。ApxIVA的存在沒(méi)能得到證實(shí)(見(jiàn)圖5)。
實(shí)施例5apxIV基因在胸膜肺炎放線桿菌感染期間的體內(nèi)表達(dá)將含有80kD多聚組氨酸-ApxIV-var3融合蛋白(見(jiàn)實(shí)施例3)的聚丙烯酰胺凝膠轉(zhuǎn)移至硝酸纖維素膜上。將該膜分成細(xì)條帶(strips)與抗血清型1的(100倍稀釋的)恢復(fù)期野生血清或以血清型1的參比菌株(Frey和Nicolet,獸醫(yī)微生物學(xué),28;61-73,1991)實(shí)驗(yàn)性感染的豬血清起反應(yīng)。用堿性磷酸酶標(biāo)記的抗兔IgG的偶聯(lián)物(Kirkegaard Perry公司,Graithersburg,Md)與NBT(4-氮藍(lán)四唑氯化物)及BCIP(5-溴-4-氯-3-吲哚基-磷酸酯)顯色觀察反應(yīng)(見(jiàn)圖6)。血清型1野生型血清及實(shí)驗(yàn)性感染的豬血清與80kD的多聚組氨酸-ApxIV-var3蛋白反應(yīng)。這表明ApxIV蛋白實(shí)際上表達(dá),具有抗原性且在豬中胸膜肺炎放線桿菌感染過(guò)程中誘導(dǎo)抗-ApxIV毒素抗體。
實(shí)施例6用Southern印跡(證明)apxIV基因在所有胸膜肺炎放線桿菌血清型中的存在及其在非胸膜肺炎放線桿菌菌株中的不存在為了研究apxIV基因在各種胸膜肺炎放線桿菌血清型及相關(guān)細(xì)菌中的存在,制備三種探針(見(jiàn)圖4)。探針APXIVA在實(shí)施例1中描述過(guò)。探針APXIVA2在BamHI和NruI位點(diǎn)之間含有2015bp的DNA片段。通過(guò)用寡聚體APPIV1-L(5’-GGGACAGTGGCTCAATTAAG-3’)和(APPIV1-R(5’-AGCTGTAAACTCCACCAACG-3’)的PCR擴(kuò)增制備758bp的探針APPIVA1。所有的探針根據(jù)制造商手冊(cè)以洋地黃毒苷-11-dUTP(寶靈曼)標(biāo)記并與Southern印跡雜交,印跡中含有所有胸膜肺炎放線桿菌參比菌株和HV114野生菌株、豬放線桿菌(ATCC15558)、羅氏放線桿菌(ATCC27072)及馬駒放線桿菌(ATCC19392)的ClaI消化的染色體DNA。所有三個(gè)探針?lè)磻?yīng)類似(見(jiàn)圖7APXIVA2探針的結(jié)果)。所有的胸膜肺炎放線桿菌菌株反應(yīng),而沒(méi)有觀察到與豬放線桿菌、馬駒放線桿菌及羅氏放線桿菌菌株的反應(yīng)。
實(shí)施例7用PCR擴(kuò)增(證實(shí))apxIV基因在胸膜肺炎放線桿菌及相關(guān)菌株中的存在用來(lái)自各種胸膜肺炎放線桿菌血清型、其它放線桿菌種類及溶血巴斯德氏菌的50ng染色體DNA作為模板,及引物APXIVA-1L(5’-TGGCACTGACGGTGATGA-3’)和APXIVA-IR(5’-GGCCATCGACTCAACCAT-3’進(jìn)行PCR擴(kuò)增。產(chǎn)物在瓊脂糖凝膠上分析以后,在所有胸膜肺炎放線桿菌樣品中觀察到442bp預(yù)期大小的產(chǎn)物,但沒(méi)有在其它放線桿菌種類中觀察到(圖8)。這表明除了實(shí)施例6中的結(jié)果外,PCR也可用于區(qū)分胸膜肺炎放線桿菌與其它放線桿菌種類。
實(shí)施例8ApxIV-var1多聚組氨酸融合蛋白的過(guò)量表達(dá)以質(zhì)粒pROK-7(見(jiàn)實(shí)施例1)作為模板且寡核苷酸APX4/II5-L(5’-CGCCATATGACAAAATTAACTATGCAAG)及APX4II6-R(5’-CGCGAATTCAGCGACACAAGAGATAT)作為PCR-引物開(kāi)始,擴(kuò)增PCR片段。然后將足夠量的該片段以限制性酶NdeI和EcoRI消化并克隆入實(shí)施例3中所述的以相同酶消化的表達(dá)載體pETHIS-1中。根據(jù)得到的命名為pJFFApxIVA1His1的質(zhì)粒,按實(shí)施例3中所述在大腸桿菌中過(guò)量表達(dá)206kD的1841個(gè)氨基酸殘基的多聚組氨酸融合蛋白(PAGE中測(cè)定的MW)。如在SEQ ID NO5中所述,該蛋白由橫跨其中no.1132至6546核酸的編碼區(qū)所編碼。蛋白的氨基酸序列顯示于SEQ ID NO6中。在Western印跡中,顯示此產(chǎn)物與特異性的抗-ApxIV血清522-409反應(yīng)(實(shí)施例3中描述的抗血清)。
實(shí)施例9通過(guò)以ApxIV接種保護(hù)小鼠免受APP感染兩種均源自血清型1參比菌株4074基因組材料的在實(shí)施例8中所述206KD多聚組氨酸-ApxIV融合蛋白和相應(yīng)的108KD多聚組氨酸-ApxIA-融合蛋白按實(shí)施例3中所述過(guò)量表達(dá)。將誘導(dǎo)大腸桿菌細(xì)胞的細(xì)胞沉淀物重懸于PBS緩沖液(1g細(xì)胞沉淀于6ml緩沖液中)并于冰上45秒鐘超聲粉碎2次用于細(xì)胞裂解。以22.000xg 4℃離心20分鐘后,棄上清并將3M的尿素(pH6.3)溶液洗得到的沉淀。22.000xg離心20分鐘后去除尿素,并將得到的沉淀溶于6M的鹽酸胍(pH8.0)中。經(jīng)PAGE凝膠的光密度測(cè)定后通過(guò)特定蛋白的濃度對(duì)蛋白樣品進(jìn)行標(biāo)準(zhǔn)化。樣品以PBS稀釋并以油性佐劑配制。
15只小鼠為一組共3組以ApxIV抗原(36.3微克)、ApxIA抗原(36.3微克)或僅以佐劑進(jìn)行腹膜內(nèi)免疫。以0.4ml體積施用疫苗。第一次接種24天后,將每組小鼠分成7只和8只小鼠的兩組,均以一半量的抗原增強(qiáng)免疫。7只小鼠的組以0.2ml的體積在腹膜內(nèi)進(jìn)行增強(qiáng)免疫且8只小鼠的組以0.1ml每只于后肢進(jìn)行肌肉內(nèi)接種。增強(qiáng)免疫13天后,將小鼠1.5108cfu的毒性血清型1菌株腹膜內(nèi)感染。
實(shí)施例10ApxIV的孔道形成能力表達(dá)ApxIV的新鮮誘導(dǎo)的大腸桿菌用作用于按Maier等,感染免疫,64;4415-4423(1996)所述在人工脂雙層中測(cè)試孔道形成的蛋白來(lái)源。用于黑色脂雙層實(shí)驗(yàn)的方法以前已有描述(Benz等;生物化學(xué)生物物理學(xué)報(bào)511305-319(1978)。膜由1%的asolection溶液(Sigma的大豆凝集素型IV-S,St,Louis MO)于正癸烷中形成。形成10倍跨膜鹽梯度后5-10分鐘用Keithley 610C靜電計(jì)測(cè)量零電流膜電勢(shì)(Zero currentmembrane potential)(Benz等,生物化學(xué)生物物理雜志551238-247(1979)。在0.5%膽固醇存在時(shí)ApxIV的存在導(dǎo)致高頻率的孔道形成,平均單個(gè)通道電導(dǎo)率(G)為4nS。
這些結(jié)果表示ApxIV誘導(dǎo)進(jìn)入人工雙層的孔道且因此對(duì)真核細(xì)胞是有毒的且因此為胸膜肺炎放線桿菌的毒性因子。
