本發(fā)明屬于病毒分子檢測技術(shù)領(lǐng)域,具體涉及一種鯽魚冠狀病毒hb93的rt-lamp檢測引物及應(yīng)用。
背景技術(shù):
國際病毒分類委員會(internationalcommitteeontaxonomyofviruses,ictv)于2012年發(fā)表了最新的病毒分類第九次報告,將套式病毒目(nidovirales)中的冠狀病毒科(coronaviridea)中新設(shè)冠狀病毒亞科(coronavirinea)和環(huán)曲病毒亞科(torovirinae)。
冠狀病毒科是一類線形單股正鏈rna病毒,最早于1937年從雞身上分離出來,almeida等對這些病毒進行了形態(tài)學(xué)研究,電子顯微鏡觀察發(fā)現(xiàn)這些病毒的包膜上有形似日冕的棘突,故提出命名這類病毒為冠狀病毒[1]。1975年,國際病毒命名和分類委員會正式命名了冠狀病毒科。2002-2003年期間,自我國局部地區(qū)發(fā)生的非典型肺炎,即由冠狀病毒屬的嚴(yán)重急性呼吸綜合征病毒(sars-cov)引起,世界上有30多個國家和地區(qū)相繼報道出現(xiàn)病例,病死率超過10%。2012年,中東地區(qū)開始出現(xiàn)新型冠狀病毒,病死率超過50%,引起了世界范圍的廣泛關(guān)注,造成社會對冠狀病毒的恐慌。因此,對冠狀病毒進行深入研究具有重要意義。
目前對冠狀病毒科的研究主要集中在冠狀病毒亞科,而對環(huán)曲病毒亞科研究甚少。其中,2006年鳊魚病毒屬(bafinivirus)的中的鳊魚病毒(whitebreamvirus)由schützeh等科學(xué)家在鳊魚體內(nèi)發(fā)現(xiàn),這也是目前ictv分類中確定的唯一以水生動物為宿主的冠狀病毒。
目前鯽魚冠狀病毒(cruciancarpbafinivirus,ccbv)尚無公開報道,更也無有效的檢測方法。
技術(shù)實現(xiàn)要素:

本發(fā)明的目的在于提供了一種鯽魚冠狀病毒hb93的rt-lamp檢測引物的應(yīng)用,所述的引物可用于制備檢測鯽魚冠狀病毒的檢測試劑盒。
為了達到上述目的,本發(fā)明采取以下技術(shù)措施:

一種鯽魚冠狀病毒hb93的rt-lamp檢測引物的應(yīng)用,包括利用上述引物制備檢測鯽魚冠狀病毒的檢測試劑盒。
優(yōu)選的,當(dāng)制備成試劑盒時,該試劑盒的非診斷性檢測方法包括:
一種鯽魚冠狀病毒hb93的rt-lamp檢測方法,步驟包括:
1)提取待測樣品的rna;
2)rt-lamp擴增:
采用25μl反應(yīng)體系:內(nèi)引物fip和bip各0.8μm,外引物f3和b3各0.1μm,dntps1mm,betaine0.5m,dtt4mm,mgcl22-10mm,bstdna聚合酶8u,amv逆轉(zhuǎn)錄酶5u,模板rna5μl,10×thermopolreactionbuffer2.5μl,選擇55-65℃和30-120分鐘進行rt-lamp反應(yīng)之后,80℃滅活2分鐘;
3)、檢測結(jié)果判定,采用下述三種方式之一:
a、發(fā)生擴增反應(yīng)時,從dntps析出的焦磷酸根離子和反應(yīng)液中的鎂離子形成白色焦磷酸鎂沉淀,通過肉眼判斷:檢測管出現(xiàn)明顯渾濁為陽性,未見渾濁為陰性;
b、每25μl體系的反應(yīng)管加1000×sybrgreeni1-2μl,1-5min觀察結(jié)果,反應(yīng)液變綠為陽性,保持橙色為陰性;
c、取擴增產(chǎn)物,用2%w/v的瓊脂糖凝膠電泳后,置于凝膠成像系統(tǒng)中成像,電泳圖片顯示lamp特征性梯狀條帶,則結(jié)果為陽性;如無任何條帶,則結(jié)果為陰性。
以上所述的方法中,優(yōu)選的,mgcl2的濃度為7mm,反應(yīng)溫度為64℃,模板rna在95℃5分鐘后置于冰浴中進行預(yù)處理。
與現(xiàn)有技術(shù)相比,本發(fā)明具有以下優(yōu)點:
1、本發(fā)明公開了一種鯽魚冠狀病毒rt-lamp檢測試劑盒;
2、特異性好,能有效檢測出ccbv病毒;
3、靈敏性好,最低檢測限可達11.48pg/μl的rna;
4、快速高效,檢測時間約1小時,非常適用于ccbv的現(xiàn)場快速檢測;
5、不需要特殊試劑與設(shè)備,檢測成本低;
6、鑒定簡單,通過從dntps析出的焦磷酸根離子和反應(yīng)液中的鎂離子形成白色焦磷酸鎂沉淀,經(jīng)肉眼就可判斷結(jié)果,通過加入1000×sybrgreeni,可提高結(jié)果的靈敏度。
具體實施方式
本發(fā)明所述技術(shù)方案,如未特別說明,均為本領(lǐng)域的常規(guī)方案,所用試劑或原料,如未特別說明,均購自商業(yè)渠道或是已公開。
實施例1:
一種鯽魚冠狀病毒hb93(本發(fā)明或稱為ccbv-hb93),其分離過程如下:
1)將攜帶病毒的鯽魚肝、腦、脾、腎等組織取出并碾磨后,培養(yǎng)基稀釋100倍,2000rpm離心后取上清過濾,接種到草魚性腺細(xì)胞(grasscarpovarycellline,gco)單層,25℃培養(yǎng)7天,產(chǎn)生細(xì)胞病變效應(yīng)(cpe),可見gco細(xì)胞收縮變圓、折光度增加,部分細(xì)胞開始脫落,細(xì)胞單層開始破裂。
培養(yǎng)基:m199,10%胎牛血清,ph7.0~7.2
2)收集病變細(xì)胞,利用透射電鏡進行形態(tài)學(xué)鑒定。
3)收集病毒,對其進行全基因組測序,序列為seqidno.1所示。
鯽魚冠狀病毒hb93的形態(tài)特征:病毒呈棒狀,四周有明顯的日冕狀棘突結(jié)構(gòu),與典型的冠狀病毒形態(tài)相符,該病毒已于2017年5月11日送至中國典型培養(yǎng)物保藏中心進行保藏,分類命名:鯽魚冠狀病毒(cruciancarpbafinivirus)hb93,保藏編號:cctccno:v201722,地址:中國武漢武漢大學(xué)。
實施例2:
一種鯽魚冠狀病毒hb93rt-lamp檢測引物,包括:

實施例3:
利用鯽魚冠狀病毒hb93rt-lamp檢測引物檢測鯽魚冠狀病毒hb93的方法,包括:

實施例4:
鯽魚冠狀病毒rt-lamp檢測引物特異性實驗
利用實施例3所述的方法,對表1中的待檢病毒樣品進行檢測,同時設(shè)置陰性對照組(雙蒸水)和陽性對照組(ccbv-hb93),待檢病毒樣品具體為:鯉春病毒血癥病毒(svcv),草魚呼腸孤病毒873(gcrv-873),草魚呼腸孤病毒9014(gcrv-9014),傳染性造血器官壞死病毒(ihnv)。

結(jié)果如表1所示,僅陽性對照組(ccbv-hb93)顯示lamp特征性梯狀條帶,其余組均無特征條帶,說明鯽魚冠狀病毒hb93rt-lamp檢測引物具備很強的特異性。
實施例5:
鯽魚冠狀病毒rt-lamp檢測引物的靈敏度
將濃度為1148ng/μl的ccbv-hb93rna模板10倍梯度稀釋,利用實施例3的方法,檢測利用本發(fā)明提供的rt-lamp引物的檢測限,105倍稀釋還可觀察到特征性梯狀條帶,也就是最低可檢測11.48pg/μl的rna模板。
