本發(fā)明涉及與玉米籽粒發(fā)育相關的13個新的bZIP轉(zhuǎn)錄因子及其編碼基因,并提 供了這些轉(zhuǎn)錄因子基因在籽粒發(fā)育過程中的表達特征。
背景技術:
玉米(&"ma"L),又稱苞米,是世界上第三大糧食作物,播種面積僅次于水稻 和小麥。從2000年起,玉米的總產(chǎn)量和單產(chǎn)量均居第一位。玉米不僅是高產(chǎn)的糧食 作物,也是營養(yǎng)豐富的詞料作物和良好的工業(yè)原料作物。玉米籽粒是玉米最主要的生 產(chǎn)性狀。除了能為人們提供糧食來源外,實際上也是重要的繁殖器官,是未本科種子 發(fā)育研究的重要模型。玉米籽粒的發(fā)育是從受精開始的,通過雙受精過程,從花粉管 尖端釋放出的一個精子與卵細胞融合,形成二倍體的合子,發(fā)育成胚;另一個精子與 兩個極核融合后發(fā)育成三倍體的胚乳。在整個發(fā)育過程中,籽粒經(jīng)過早期胚胎發(fā)生、 成熟以及休眠,然后在適當?shù)臏囟群退輻l件下,開始萌發(fā)。
從分子生物學的角度來說,籽粒的發(fā)育和萌發(fā)是一個有次序的、選擇性的基因表 達過程,而轉(zhuǎn)錄因子在這一過程中起到了關鍵性的作用。轉(zhuǎn)錄因子(transcription factor) 也稱為反式作用因子(trans-actingfactor),是一類能夠與真核基因啟動子區(qū)順式作用元 件(cis-acting factor)特異性結合的DNA結合蛋白。通過轉(zhuǎn)錄因子與順式作用元件之間 以及它們和其他相關蛋白之間的相互作用,轉(zhuǎn)錄因子能夠激活或抑制下游基因的轉(zhuǎn) 錄。根據(jù)轉(zhuǎn)錄因子DNA結合域的保守性,典型的植物轉(zhuǎn)錄因子常包括bZIP結構域、 鋅指(Zinc-Finger)結構域、MADS結構域、MYC結構域、MYB結構域、Homeo 結構域、bHLH (堿性螺旋-環(huán)-螺旋)結構域及AP2/EREBP結構域等。
堿性亮氨酸拉鏈(basic leucine zipper,bZIP)蛋白是真核生物的轉(zhuǎn)錄因子中最大 且最保守的一類轉(zhuǎn)錄因子,該類轉(zhuǎn)錄因子的共同特點是(1)含有與特異DNA序列 相結合的堿性結構域;(2)參與寡聚化作用的亮氨酸拉鏈區(qū)與堿性區(qū)緊密相連;(3) 轉(zhuǎn)錄因子的N-末端含有酸性激活區(qū);(4)以二聚體的形式結合DNA,肽鏈N-末端的堿 性區(qū)與DNA直接結合。在高等植物中,bZIP蛋白與種子貯藏基因表達、光形態(tài)發(fā)生 及器官建成的控制相關。
在玉米籽粒中,關于bZIP類轉(zhuǎn)錄因子的研究相對較少。作為玉米籽粒中研究的最 透徹的一種編碼bZIP轉(zhuǎn)錄因子的基因,Opaque2((92)基因不僅調(diào)控玉米重要的貯藏蛋 白,如a-醇溶蛋白的特異性表達,同時還調(diào)節(jié)玉米6-馬cy尸尸Z)A7基因的表達。此外, 玉米中其它的bZIP轉(zhuǎn)錄因子也有少量研究,如05F3./和05F3.2與小麥的HBP-lb同 源,在種子萌發(fā)的初期表達,調(diào)控細胞周期相關的組氨酸基因(Rhonda等,1993; Tetsuya等,1991);與小麥EmBP-l同源的mEmBP-l雖然在胚乳中表達量很低,但仍 能識別22kDaa-醇溶蛋白啟動子區(qū)域的02-6ox(Leslie等,2004);在種子脫水過程中, v4S/能調(diào)節(jié)ABA應答基因的表達(Niu等,2002)。這些研究都表明,bZIP轉(zhuǎn)錄因子 對高等植物基因表達與調(diào)控十分重要。
然而,在玉米中,特別是玉米籽粒中,仍有大量的bZIP轉(zhuǎn)錄因子還沒有被發(fā)現(xiàn) 和深入研究。新的轉(zhuǎn)錄因子的發(fā)現(xiàn)和研究,將對基因工程改造玉米籽粒性狀提供重要 幫助。隨著模式植物擬南芥和水稻基因組的測序完成,人們對轉(zhuǎn)錄因子的研究已經(jīng)從 單個基因?qū)用孓D(zhuǎn)移到了基因組層面。Riechmann(2000)的研究表明在擬南芥基因組中 約有6%的基因是編碼轉(zhuǎn)錄因子的,Duan(2005)的研究也顯示在水稻種子中特異表達 的轉(zhuǎn)錄因子有135個。為解析玉米籽粒發(fā)育過程中的基因表達調(diào)控,以及這些轉(zhuǎn)錄因 子的基因工程應用提供了基礎。
發(fā)明內(nèi)容
本發(fā)明的目的之一在于提供與玉米籽粒發(fā)育相關的13個新的bZIP類轉(zhuǎn)錄因子,
命名為MSBZIP (Maize Seed bZIP) 1-13。
本發(fā)明的目的之二在于提供該轉(zhuǎn)錄因子的編碼基因。
本發(fā)明的目的之三在于提供該轉(zhuǎn)錄因子在玉米籽粒發(fā)育過程中的表達。
為達到上述目的,本發(fā)明借助基因組學研究思路,采用了玉米籽粒全發(fā)育階段的
轉(zhuǎn)錄組學分析,系統(tǒng)發(fā)掘了在籽粒發(fā)育過程中特異表達的轉(zhuǎn)錄因子,總共提供了 13
個新的bZIP轉(zhuǎn)錄因子基因,并對它們的表達特征進行了分析。
本發(fā)明采用如下的技術方案
與玉米籽粒發(fā)育相關的bZIP轉(zhuǎn)錄因子MSBZIP1-13,其特征在于該轉(zhuǎn)錄因子是 具有序列表中序列14-26的氨基酸殘基序列的蛋白質(zhì),或者是將序列14-26的氨基 酸殘基序列經(jīng)過一個或幾個氨基酸殘基的取代、缺失或添加且具有與序列14-26 的氨基酸殘基序列相同活性的由序列14-26衍生的蛋白質(zhì)。
上述的轉(zhuǎn)錄因子是具有序列表中序列14-26的氨基酸殘基序列的蛋白質(zhì)。
上述的與玉米籽粒發(fā)育相關的bZIP轉(zhuǎn)錄因子MSBZIP1-13的編碼基因,其特 征在于該編碼基因具有下列核苷酸序列之一
1) 序列表中序列1-13的DNA序列;
2) 與序列表中序列1-13限定的核酸序列具有90%以上的同源性,且編碼相同 功能蛋白質(zhì)的DNA序列。
上述的編碼基因具有序列表中序列1-13的DNA序列。
上述的與玉米籽粒發(fā)育相關的bZIP轉(zhuǎn)錄因子MSBZIP1-13的表達,其序列特征 在于該轉(zhuǎn)錄因子中MSBZIP2、MSBZIP4、MSBZIP6、MSBZIP7、MSBZIP8、MSBZIP10、 MSBZIPll、 MSBZIP12以及MSBZIP13在籽粒發(fā)育過程中持續(xù)表達的;而 MSBZIP1、 MSBZIP3、 MSBZIP5、 MSBZIP9則是在發(fā)育過程中呈階段特異性表達。
本發(fā)明提供了 13個編碼在玉米籽粒發(fā)育過程中表達的bZIP轉(zhuǎn)錄因子新基因,并 對它們進行了進一步的分析,按所含保守區(qū)段的差異可將這13個bZIP蛋白分為三大 類,它們均含有保守的堿性亮氨酸區(qū)段,但在進化上呈現(xiàn)兩種不同的模式,參見圖2、 圖3。半定量PCR分析,參見圖4,表明大多數(shù)編碼bZIP轉(zhuǎn)錄因子的基因在籽粒 發(fā)育過程中是持續(xù)表達的,在表達上呈現(xiàn)階段特異性的僅4個(約占1/3),推測特異 的表達區(qū)間可能與玉米籽粒發(fā)育的發(fā)育進程相一致,這為今后進一步研究玉米籽粒的 表達調(diào)控網(wǎng)絡奠定了基礎。
圖1顯示了對13個新基因所作的blastx和pfam分析。其中兩者的參數(shù)都設為E value〈l(T10。
圖2顯示了對bZIP轉(zhuǎn)錄因子的保守區(qū)段的分析。其中紅色方格代表保守的堿性 亮氨酸區(qū)域,黃色和綠色分別代表保守的未知功能區(qū)段。
圖3顯示了運用Megabase分析軟件對bZIP轉(zhuǎn)錄因子作的聚類分析。其中02 作為聚類分析的根。
圖4顯示了對bZIP轉(zhuǎn)錄因子在整個發(fā)育階段的表達譜分析。泳道1-14:分別是 授粉后3、 5、 7、 9、 11、 13、 15、 18、 21、 24、 27、 30、 33、 36天的不同bZIP轉(zhuǎn)錄 因子的RT-PCR; cGAPDH作為內(nèi)參。
具體實施例方式
下面結合具體實施例,進一步闡述本發(fā)明。應理解,這些實例僅用于說明本發(fā)明 而不用于限制本發(fā)明的范圍。下列實施例中未注明具體實驗條件的實驗方法,通常按 照常規(guī)條件,如分子克隆(Molecular Cloning:A Laboratory Manual,3rded.)或植物分 子生物學一實驗手冊(Plant Molecular Biology—A Laboratory Manual, Melody S. Clark 編,Springer-verlag Berlin Heidelberg, 1997)中所述條件,或按照制造廠商所建議的 條件。
實施例一玉米籽粒發(fā)育過程中表達的bZIP轉(zhuǎn)錄因子新基因的獲得
1. 玉米籽粒發(fā)育階段均一化cDNA文庫的構建
為了得到整個籽粒發(fā)育階段的表達序列,收集授粉后3天-36天的玉米籽粒,將 它們等體積混合后,提取總RNA (Trizol kit, Qiagen),以經(jīng)Promega公司的PolyA TtractR mRNA Isolation System III Kit純化所得的mRNA為模板,利用基于復性動力 學的原理的cDNA均一化方案,即通過變性一復性操作,使cDNA的豐度趨于一致, 最終建立均一化的cDNA文庫。
2. 編碼bZIP轉(zhuǎn)錄因子基因的獲得
采用測序通用引物T3/T7 ,雙脫氧法(DYEnamic ET-Terminator Cycle Sequencing), DNA序列分析儀(MegaBACE 4500)對隨機挑選的10,000個克隆進行 測序。所得序列經(jīng)PhredPhrap程序組分析,得到代表籽粒發(fā)育階段表達的EST文庫。 所得序歹U經(jīng) blastx( http:〃www.ncbi.nlm.nih.gov/dbEST)禾卩 pfam(http:〃www. pfam.jouy.inra.fr)分析,共得到13個編碼bZIP轉(zhuǎn)錄因子的新基因,參見圖1,其中除 4個與水稻中已知功能bZIP轉(zhuǎn)錄因子同源外,其余都是9個均為功能未知的新的bZIP 蛋白,我們將其命名為MSBZIP (Maize Seed bZIP) 1-13。。
實施例二 bZIP轉(zhuǎn)錄因子的鑒定
運用MEME分析軟件對這13個bZIP轉(zhuǎn)錄因子進行保守區(qū)段分析,參見如圖2, 它們都含有一個保守的堿性亮氨酸區(qū)域,而MSBZIP7-12除了含有bZIP蛋白特有的 堿性區(qū)段外,還具有一個功能未知的保守區(qū)段。根據(jù)所含保守區(qū)段的個數(shù)和種類,我 們將這13個轉(zhuǎn)錄因子分為3類,其中MSBZIP1-6屬于第一類;MSBZIP7-11歸為第 二類;而第三類則包括MSBZIP12和MSBZIP13。
利用Megabase3.0軟件對這13個bZIP轉(zhuǎn)錄因子進行聚類分析,參見圖3,結果 顯示MSBZIP5-11在進化距離上比較接近,而MSBZIPl-4、 MSBZIP12、 MSBZIP13 以及6>2在距離上較接近,這可能揭示了存在于這些bZIP轉(zhuǎn)錄因子中的兩種進化模 式。
實施例三bZIP轉(zhuǎn)錄因子的表達譜分析
以授粉后3-36天的玉米籽粒RNA為模板,Oligo (dT) 16作引物進行反轉(zhuǎn)錄, 得到整個籽粒發(fā)育階段各個時間點的cDNA。根據(jù)這13個轉(zhuǎn)錄因子的序列分別設計 引物,參見表l,以各個時間點的cDNA為模板進行半定量PCR, PCR產(chǎn)物經(jīng)1X瓊 脂糖凝膠電泳分析,參見圖4,證明MSBZIP2、 MSBZIP4、 MSBZIP6、 MSBZIP7、 MSBZIP8、 MSBZIPIO、 MSBZIPll、 MSBZIP12以及MSBZIP13都是在籽粒發(fā)育過 程中持續(xù)表達,參見圖4-A;而MSBZIP1、 MSBZIP3、 MSBZIP5、 MSBZIP9則是在 發(fā)育過程中特異性表達,參見圖4-B。

本發(fā)明涉及到13個在玉米籽粒發(fā)育過程中表達的bZIP轉(zhuǎn)錄因子新基因以及對它們的序列分析和表達分析。本發(fā)明提供的13個bZIP類轉(zhuǎn)錄因子基因名稱為MSBZIP1-MSBZIP13,其核酸序列是下列核苷酸序列之一1)序列表中1-13的DNA序列;2)與序列表中序列1-13限定的DNA序列有90%以上同源性,且編碼相同功能蛋白質(zhì)的DNA序列。bZIP類轉(zhuǎn)錄因子基因所編碼的蛋白具有序列表中序列14-26的氨基酸殘基序列,或者是將序列14-26的氨基酸殘基序列經(jīng)過一個或幾個氨基酸殘基的取代、缺失或添加且具有與序列14-26的氨基酸殘基序列相同活性的由序列14-26衍生的蛋白質(zhì)序列。本發(fā)明所公開的這13個bZIP轉(zhuǎn)錄因子基因是在玉米中首次報道的,這為解析玉米籽粒發(fā)育過程中的基因表達調(diào)控,以及這些轉(zhuǎn)錄因子的基因工程應用提供了基礎。
